File type ANME is an Anime Studio file. You can ONLY open it in Anime Studio. What do you want to open the file with? If you are expecting to open a movie file then you must export the animation as a movie.

You didn't say what kind of file it is. If it's an image file then just create a new image layer and give it the name of the file.

Brushes only exist in the PRO version. The basic version doesn't have them. I asked the same question a month ago when I first got started. Seems the tutorials don't really make that clear.

They neglect to mention that File > Render is in the PRO version only, not the basic version. To see the rendered result in non-PRO version you have to use File > Export Animation. Are you SELECTING the object before you change the color? Otherwise, you will just be changing he default color, not the color of the object you've selected. Does the object you select show the checkerboard pattern to show that it is selected?
