Seems to me the only things you truly need are a pencil, paper and possibly some inks if you want to draw by hand.

If you want to do everything in the computer I would suggest a WACOM Tablet or a WACOM Cintiq and a program like Manga Studio or Photoshop.

Very well done art and style.

Only critique I have (good or bad, your choice) is that it definitely has the look of something done in Anime Studio. You can almost see the Bones at work in a lot of the movements of the characters.

Other than that it looks pretty awesome so far.

Very cool stuff.

Definitely some of the better work I've seen done in AS so far. You can definitely tell its done in AS...w/ the bouncy noses and arms and such.

I would say just learn AS and its drawing tools instead of doing double duty in Illustrator and AS. Any wacom tablet will do, personally I prefer the A5 size, too. For bitmap animation I use my Cintiq. I wish I had the money for a Cintiq. I find running around the garden helps a lot. If that's not for you, perhaps reading the tutorials and actually animating your story might work. Based on your character tests and the entire animatic I think this is going to be a great short film. I can't wait to see it finished! It's nice to see some people really show what AS can do in the right hands.

I feel like the walk is less of a waddle now...and I liked the waddle. I think having the shoulders/head dip further down would help give this walk some more personality.
